The main purpose why an inoculating loop is flamed is to kill microorganism in order to avoid contamination

    2. How will you know when you have flamed the loop or needle long enough?

It is expedient to flame the loop or needle until it is red hot, this is a temperature any microorganism can not survive

    3. Why is it necessary to cool the inoculating loop prior to obtaining the bacterial sample?

It is pertinent to cool the inoculating loop in order to avoid bacteriocidal i. e the killing of bacteria that one intended to inoculate.

    4. In which direction should you move the inoculating loop in the Bunsen burner flame? (i. e. from the handle to the loop or from the loop to the handle)

Loop must be carefully handled in order to avoid re-contamination after it has been flamed, this is why handling plays a vital role. it should be handled from the handle to the loop. This is also a precaution method in order not get the handler injured as well as to also avoid contamination
