The Meselson and Stahl experiment begin with the use of E. coli that has 15 N / 15 N marked DNA grown in 14 N media. Meselson and Stahl noted that the first generation has hybrid 15N/14N DNA, and the second generation possess hybrid 15N/14N DNA and 14N/14N DNA. No 15N/15N DNA was seen in the course of the experiment. The experiment was carried with the use of sedimentation equilibrium centrifugation to give a strong background for the semiconservative model of DNA replication.
