What is difficult to determine in the process of adaptation unless there is a closely related species to which you can compare findings?

Assuming that you're taking about animals adapting to new habitats, animals don't talk so it would be hard for a human to tell if the animal successfully adapted to it's new environment. Although we can't understand the animals, they communicate with each other.
