The process of annealing temperature varies greatly with the different PCRs this is because the type of primer is specific to the temperature.

Different set of primers have different temperature by which the DNA binds. The temperature 42°C was used on the primers like T11CG, T11GG, T11GC and T1CC and there was less complimentary fragments.

When the temperature was taken 40°C showed 2 mole folds and better complementary fragments was seen.

This concludes that annealing temperature varies with Different PCRs.
