The best answer for this question would be:

That the main theme of the book is about hope, hope for the people and the hope for change. Though, a lot of catastrophes had happened, it is not too late to change one's fate, it is not too late to hope.

In The Diary of Anne Frank, the characters feel sadness after reading the hopeful message written on the cake. The message was "Peace 1944." After reading the message everyone felt sad and worried about the life they are living. Miep gets reminded by Mrs. Van Daan about last year's cake which read "Peace in 1943." This suggests the crave desire of the people about to live in peace and harmony in their land. The people had expected peace in the year 1943 which they left deprived of. Hence, they feel terrified about the coming 1944.
