    Any change in nucleotide sequence may lead to mutations and thereby different types of disorders or diseases. Sickle cell, Tay Schs are some diseases caused by the change in nucleotide. A nucleotide is responsible for coding for a protein. So change will lead to failure of production of particular protein and cause for disease.
