What is the difference between streak plate and spread plate technique?

    For a streak plate method, you simply streak the sample on one edge of a plate by using a swab, or a sterile if you have a liquid or slurry sample. Then, you take another sample from the first part and streak it to another part of the plate. Then, streak again to another part of the plate until you have four parts of streaked sample. This would represent four dilutions. For a spread plate technique, you would put drops of sample on a plate and you spread it over the plate with the use of a sterile loop. For a streak plate, the sample is from a slant while for a spread plate, the sample is from a broth. The purpose of streak plate is to isolate colonies while, for spread plate, you are aiming to count the number of colonies.
